Control of Gluconeogenesis and of Enzymes of Glycogen Metabolism in Isolated Rat Ilepatocytes

چکیده
Hepatocytes isolated from the livers of fed rats were used for a cQmparative study of the effects of phenylephrine, vasopressin and glucagon on gluconeogenesis and Qn enzymes of glycogen metabolism. When hepatocytes were incubated in the presence of Ca2 , phanylephrine stimulated gluconeogenesis from pyruvate less than did glucagon, but, in contrast with this hormone, it did not affect the activities of protein kinase and pyruvate kintse, nor the concentration of phosphoenolpyruvate, and it did not decrease the release of 3H20 from [6-3H]glucose. The effects of vasopressin were similar to those of phenylephrine. Gluconeogenesis from fructose was also stimulated by phenylephrine and, more markedly, by glucagon at the expense ofthe conversion of fructose into lactate. Insulin was able to antagonize the stim'ulatory effect of phenylephrine on gluconeogenesis from pyruvate. When Ca2+ was removqd from the incubation medium, phenylephrine still stimulated gluconeogenesis from pyruvate, but it also caused an activation of protein kinase and an inactivation of pyruvate kinase; accordingly, the concentration ofphosphoenolpyruvate was increased, and, in contrast, vasopressin had no effect on all these parameters. The property of phenylephrine to cause the activation of glycogen phosphorylase was decreased by glucose or by the absence of Ca2+; it was abolished when these two conditions were combined. Glycogen synthase was inactivated by phenylephrine in the presence or the absence of Ca2+, although presumably by different mechanisms.

Aloe Vera contains phenolic compounds and flavonoids with antioxidant properties that reduces the complications of the disease. The aim of study was to investigate the effects of hydroalcoholic extract of Aloe Vera in serum levels of...
متن کاملBiochemical changes in carbohydrate metabolism of the fish – Cyprinus carpio during sub-lethal exposure to biopesticide – Derisom.
The effect of biopesticide - Derisom on certain metabolites and enzymes of carbohydrate metabolism were evaluated in gill, liver, kidney and muscle of Cyprinus carpio during sub-lethal toxicity exposure of 21 days. A dose of 0.28 ppm was taken as the sub-lethal dose. The organs were taken from exposed and control fish at the end of 24 hrs, 7, 14 and 21 days and used for the estimation of total ...
متن کاملNon-Classical Gluconeogenesis-Dependent Glucose Metabolism in Rhipicephalus microplus Embryonic Cell Line BME26
In this work we evaluated several genes involved in gluconeogenesis, glycolysis and glycogen metabolism, the major pathways for carbohydrate catabolism and anabolism, in the BME26 Rhipicephalus microplus embryonic cell line. Genetic and catalytic control of the genes and enzymes associated with these pathways are modulated by alterations in energy resource availability (primarily glucose). BME2...

Epinephrine and the a-adrenergic agonist phenylephrine activated phosphorylase, glycogenolysis, and gluconeogenesis from lactate in a dose-dependent manner in isolated rat liver parenchymal cells. The half-maximally active dose of epinephrine was lo-’ M and of phenylephrine was 10m6 M. These effects were blocked by a-adrenergic antagonists including phenoxybenzamine, but were largely unaffected...
متن کاملHormonal control of glycogenolysis and gluconeogenesis in isolated rat liver cells.
The carbohydrate metabolism of isolated rat liver parenchymal cells was investigated. Cells isolated from fed rats retained glycogen and released glucose into the suspending medium at a rate of about 25 pg of glucose per mg of cell protein per hour. Glucagon, epinephrine, or cyclic nucleotides stimulated glycogenolysis about 220 to 250%. Cells isolated from rats fasted 18 to 24 hours synthesize...
